    Reconsideration of In-Silico siRNA Design Based on Feature Selection: A Cross-Platform Data Integration Perspective

    RNA interference via exogenous short interference RNAs (siRNA) is increasingly more widely employed as a tool in gene function studies, drug target discovery and disease treatment. Currently there is a strong need for rational siRNA design to achieve more reliable and specific gene silencing; and to keep up with the increasing needs for a wider range of applications. While progress has been made in the ability to design siRNAs with specific targets, we are clearly at an infancy stage towards achieving rational design of siRNAs with high efficacy. Among the many obstacles to overcome, lack of general understanding of what sequence features of siRNAs may affect their silencing efficacy and of large-scale homogeneous data needed to carry out such association analyses represents two challenges. To address these issues, we investigated a feature-selection based in-silico siRNA design from a novel cross-platform data integration perspective. An integration analysis of 4,482 siRNAs from ten meta-datasets was conducted for ranking siRNA features, according to their possible importance to the silencing efficacy of siRNAs across heterogeneous data sources. Our ranking analysis revealed for the first time the most relevant features based on cross-platform experiments, which compares favorably with the traditional in-silico siRNA feature screening based on the small samples of individual platform data. We believe that our feature ranking analysis can offer more creditable suggestions to help improving the design of siRNA with specific silencing targets.     Diplomacia cultural : a promoção cultural dos consulados-gerais em Porto Alegre

    Esta pesquisa tem como tema central a Diplomacia Cultural, buscando entender a partir dela como os Consulados-Gerais presentes na cidade de Porto Alegre trabalham comunicacionalmente a cultura de seus países através da promoção de atividades culturais. A escolha do tema foi feita partir da busca por entender melhor as culturas de diferentes países e como elas se disseminam ao redor do mundo. A partir disso definiu-se como objetivo geral desta monografia analisar se a Diplomacia Cultural exercida pelos Consulados-Gerais da Argentina, Chile, Espanha, Itália e Uruguai, presentes na cidade de Porto Alegre no Estado do Rio Grande do Sul, é efetiva. Utilizou-se como principais autores teóricos Ribeiro (1989), apresentando a Diplomacia Cultural, Nobre (2016), apresentado a relação das Relações Públicas com a diplomacia e Rubin (2005), apresentado o Sistema Cultural a ser utilizado posteriormente na análise. Foi utilizado como metodologia de pesquisa o Estudo de Caso a partir da visão de Yin (2001), que inclui as técnicas de pesquisa bibliográfica e análise de conteúdo. A partir da análise dos resultados obtidos foi possível entender como cada país trabalha a diplomacia cultural no estado do Rio Grande do Sul, destacando se essa diplomacia cultural é de fato efetiva, apresentando as razões que fazem com que esses países trabalhem a cultura de determinada forma na região e apontando novas perspectivas acerca do tema escolhido.This research has, as a main topic, the cultural diplomacy, seeking to understand how the general consulates existents in Porto Alegre works communicationally the culture of their countries through the promotion of cultural activities. The choice of the theme was made in the quest to understrand better the cultures of different countries and how they are dissiminated all around the world. From this, was defined, as the main objective of this research, to analyse if the Cultural Diplomacy exercised by the General Consulates of Argentina, Chile, Spain, Italy and Uruguay, presents in the city of Porto Alegre, State of Rio Grande do Sul, are efective. The main authors used was Ribeiro (1989) – presenting the Cultural Diplomacy – Nobre (2016) – showing the Public Relationships with diplomacy – and Rubin (2005), showing the Cultural System to be used after the analysis. It was used, as methodology, the case study, from the vision of Yin (2001), that included the technics of bibliographic research and content analysis. From the obtained results, it was possible to understand how each country works with cultural diplomacy in the state of Rio Grande do Sul, highlighting if this cultural diplomacy is, in fact, efective, showing the reasons that made these countries works with the culture of certain shape in region and pointing new perspectives about the chosen topic
